Glycoxidative stress creates a vicious cycle of neurodegeneration in Alzheimer's disease--a target for neuroprotective treatment strategies?

Accumulation of Advanced Glycation Endproducts (AGEs) in the brain is a feature of ageing and degeneration, especially in Alzheimer's disease (AD). Increased AGE levels explain many of the neuropathological and biochemical features of AD such as extensive protein crosslinking (beta-amyloid and MAP-tau), glial activation, oxidative stress and neuronal cell death. Oxidative stress and AGEs initiate a positive feedback loop, where normal age-related changes develop into a pathophysiological cascade. Combined intervention using antioxidants, anti-inflammatory drugs and AGE-inhibitors may be a promising neuroprotective strategy.

El estrés glicosilativo crea un círculo vicioso de neurodegeneración en la enfermedad de Alzheimer. Un blanco estratégico para la neuroprotección?
La acumulación de productos de glicación avanzados(AGEs). en el cerebro es una de las característicasdel envejecimiento y la degeneración especialmente en la enfermedad de Alzheimer. La elevación de los niveles de AGEs explican muchas de las características neurofisiopatológicas y bioquímicas de de la enfemedad de Alzheimer tales como el extenso proceso de ligadura entre proteinas glicadas y productos amiloides , activación de la glía, estrés oxidativo y muestre neuronal. El estrés oxidativo y los producto AGEs inician un círculo vicioso donde los cambios relacionados con la edad entran en una cascada fisiopatológica. La intervensión combinadausando antioxidantes, antiinflamatorios e inhibidores de los AGEs pueden ser una estrategia neuroprotectora.
